- JAK Inhibition - Tofacitinib is classified as a Janus kinase inhibitor (JAK inhibitor). The JAK family includes four enzymes: JAK1, JAK2, JAK3, and TYK2. These enzymes play a critical role in transmitting signals from cytokine receptors on the surface of cells to the inside of the cell, influencing gene expression and immune cell function.
